Title: Characterization of gut bacteria of the Italian honey bee Apis mellifera (Apidae: Hymenoptera)
Objectives & Work Carried Out:
- Profiling the diversity and composition of gut bacterial communities in A. mellifera using both culture-dependent and culture-independent approaches.
- Isolation and culturing of gut microorganisms followed by DNA extraction, PCR, ARDRA, RFLP, and PLFA analysis.
- Metagenomic sequencing and downstream analysis including alpha and beta diversity, phylogenetic analysis, CAZy (Carbohydrate-Active Enzyme) profiling, and functional pathway analysis.
- GC–MS analysis for metabolic profiling of gut bacterial communities.
- Functional assessment of selected gut bacterial isolates through qualitative and quantitative enzyme assays.
- Estimation of cellulase, hemicellulase, pectinase, lipase, and invertase activities using spectrophotometric methods.

Title: Genome-wide identification, characterization, expression, and functional analysis of odorant-binding proteins and chemosensory proteins in the whitefly, Bemisia tabaci
Objectives & Work Carried Out:
- Genome-wide identification and annotation of odorant-binding proteins (OBPs) and chemosensory proteins (CSPs) across B. tabaci genetic groups.
- Full-length gene characterization, chromosomal localization, and comparative genomic analysis.
- Stage- and tissue-specific expression profiling of selected OBP/CSP genes using RT-qPCR.
- Protein characterization, including sequence analysis, motif identification, structural modeling, and in-silico ligand docking.
- Functional validation through RNA interference (RNAi)-mediated gene silencing and downstream phenotypic assays.
- Gene cloning, recombinant protein expression, SDS-PAGE, and ligand-binding assays.
- Behavioral validation using olfactometer assays and oviposition bioassays.
- Insecticide resistance screening and evaluation of chemosensory involvement.
- Development of attractant-based traps and implementation in large-scale field trials.
